for "speaking of", I think it means the same as "I don't know what you are talking about"

Perhaps  the first  expression  is  a  little  bit  stronger  in  tone?
"can't  understand  you  have  been  speaking  of "  would  be  more  likely  to  indicate  the  speaker's  slight  or  snobbish  air.  He  didn't  lean  a  willing  ear  to  YOUR  words  or  he  thought  it's  too  naive  to  worth  a  listening.
"can't  understand  you  have  been  saying  of " , on  the  other  hand,  possibly  conveyed  the  speaker's  puzzle  or  misunderstanding  upon  YOUR  idea.  He  failed  to  follow  ur  train  thought,  and  maybe,  u'd  better  change  an  easier  way  of  expression.

To my thinking, 'speaking of 'gives emphasis to the content you referred to, and '' saying of' leans towards the way  of your expression!!
